ENHANCING PROSTATE CANCER TREATMENT OUTCOMES: THE ROLE OF PHYSICAL ACTIVITY IN THE EFFICACY OF KANGFU XIAOYAN SUPPOSITORY

Abstract

Objective: This study aims to explore the pharmacological mechanisms of the Kangfu Xiaoyan Suppository in treating prostate cancer (PC), utilizing network pharmacology and molecular docking, and to hypothesize the potential enhancements in treatment efficacy through physical activity. Methods: Active ingredients and their corresponding targets for Kangfu Xiaoyan Suppository were identified via TCMSP, UNIPROT, Huayuan network, and Drug bank, supplemented with literature reviews. Prostate cancer targets were compiled from the GeneCards database. Common targets between the drug ingredients and disease were analyzed using Venny 2.1.0, and further investigated through STRING for protein interaction networks. Pathway enrichment analyses were conducted using the DAVID platform, focusing on involvement in prostate cancer pathways and other relevant signaling pathways like PI3K-Akt. Molecular docking of key targets with drug components was performed using Pymol and AutoDock Tools. Results: Analysis identified 315 targets from 150 active components, intersecting with 203 prostate cancer targets to highlight 8 common targets including AR, EGFR, AKT1, TP53, and GSTP1. Enrichment analysis linked these targets to critical pathways in prostate cancer development and progression. Additionally, preliminary hypotheses suggest that integrating physical activity may amplify the drug's effects, potentially through enhanced drug uptake or modulated pathway activities. Conclusion: The Kangfu Xiaoyan Suppository demonstrates potential as an effective prostate cancer treatment, with its efficacy potentially boosted by physical activity. This synergy could offer a novel approach in clinical settings, meriting further investigation into how exercise could be systematically integrated into treatment regimens for prostate cancer.
