EFFECTS OF LIVER DREDGING, SPLEEN STRENGTHENING AND HEAT CLEARING FORMULA ON PENTAZOCINE SIGNALING SYSTEM AND RELATED METABOLITES IN RATS WITH FUNCTIONAL DYSPEPSIA MODEL

Objective: To observe the effects of the formula of sparing the liver, strengthening the spleen and clearing heat on the pentraxin signaling system and related metabolites in functional dyspepsia (FD). Methods: Forty SD rats were divided into blank group (group N), model group (group M), western medicine group (group XM) and traditional Chinese medicine group (group TM) according to the random number table method, and the rats in group N were kept naturally after successful modeling, and each treatment group started treatment: rats in group M were given saline gavage, rats in group XM were given mosapride gavage, and rats in group TM were given liver-sparing and spleen-clearing formula gavage. The changes of body weight of rats in each group were observed, the morphology of gastric tissue was observed by HE staining, and the changes of 5-HT and 5-HT1A receptor protein expression and plasma metabolites in hippocampus of rats were detected by immunohistochemistry. Results: After modeling, the body weight of rats in each group decreased compared with rats in group N, and the difference was statistically significant (p<0.05); after treatment for 2W, the body weight of rats in TM and XM groups increased significantly compared with rats in group M, and the difference was statistically significant (p<0.05); the mucosal morphology of gastric sinus was intact; compared with rats in group N, the expression of 5-HT in hippocampal tissue of rats in group M increased significantly, and the difference was statistically significant (p<0.05). The difference was statistically significant (P<0.01), and the difference was not statistically significant (P>0.05); compared with the M group, the 5-HT expression in the hippocampal tissue of rats in each treatment group was significantly decreased, and the difference was statistically significant (P<0.01); compared with the N group, the 5-HT1A receptor expression in the hippocampal tissue of rats in the M group was statistically significant (P<0.01). The difference was statistically significant (P<0.01); the 5-HT1A receptor expression in hippocampal tissue of rats in the XM group was also increased, and the difference was statistically significant (P<0.05); the 5-HT1A receptor expression in hippocampal tissue of rats in the TM group was not significantly increased, and the difference was not statistically significant (P>0.05); compared with the M Compared with group M, the expression of 5-HT1A receptors in the hippocampal tissue of rats in TM group decreased significantly, and the difference was statistically significant (P<0.01), and the expression of 5-HT1A receptors in the hippocampal tissue of rats in XM group decreased, and the difference was statistically significant (P<0.05); compared with group N, a total of 29 metabolites were significantly changed in group M, and 13 identical The three metabolites of amino acid, glutamic acid and goose deoxycholic acid were found to be closely related to the 5-HT signaling system. Conclusion: The mechanism of action of the liver-sparing and spleen-clearing formula on FD, may be related to the regulation of 5-HT signaling system and phase tube metabolites.
